Kaspaza-14
Kaspaza-14 jest enzim koji je kod ljudi kodiran genom CASP14 sa hromosoma 19.[5][6][7]
Funkcija
[uredi | uredi izvor]Gen CASP14 kodira člana porodice cistein-aspartanih proteaza (kaspaza). Sekvencna aktivacija kaspaza ima centralnu ulogu u fazi izvršenja ćelijske apoptoze. Kaspaze postoje kao neaktivni proenzimi koji se podvrgavaju proteolitskoj obradi na konzerviranim asparaginskim ostacima kako bi se dobile dvije podjedinice, veliku i malu, koje se dimeriziraju i formiraju aktivni enzim. Pokazalo se da je ova kaspaza obrađena i aktivirana pomoću kaspaze-8 i 10 in vitro, i anti-Fas agonističkog antitijela ili TNF-ove apoptoze koja indukuje ligand in vivo. Ekspresija i obrada ove kaspaze može biti uključena u keratinocitnu terminalnu diferencijaciju, što je važno za formiranje kožne barijere.[7]
Prema Atlasu ljudskih proteina,[8] CASP14 proteinom je obogaćena ljudska koža i uglavnom je eksprimiran u gornjim slojevima epiderme. Prema Atlasu ćelija, protein je uglavnom lokalizovan u citosolu.[9]
