Rho GTPaze imaju fundamentalnu ulogu u brojnim ćelijskim procesima koji su inicirani vanćelijskim stimulansima koji djeluju preko receptora vezanih za G-protein. Kodirani protein može formirati kompleks s G-proteinima i stimulirati signale zavisne o rho.[7]

Pokazalo se da ARHGEF2 ima interakciju sa PAK1.[9]
