RPS27
40S ribosomni protein S27, poznat i kao metalopan-stimulin 1 ili MPS-1, jest protein koji je kod ljudi kodiran genom RPS27 sa hromosoma 1.[3][4][5] Metalopanstimulin je cinkoprstni protein za koji se pretpostavlja da je uključen popravak DNK, kao i onkogenezu.[6]

Dužina polipeptidnog lanca je 84 aminokiseline, a molekulska težina 9461 Da.[7]

Ribosomi, organele koje kataliziraju sintezu proteina, sastoje se od male podjedinice 40S i velike podjedinice 60S. Ove podjedinice zajedno sastoje se od 4 tipa RNK i približno 80 strukturno različitih proteina. Ovaj gen kodira ribosomski protein koji je komponenta podjedinice 40S. Protein pripada porodici ribosomskih proteina S27E. Sadrži domen C4 tipa cinkovog prsta koji se može vezati za cink. Pokazalo se da se kodirani protein može vezati za nukleinske kiseline. Nalazi se u citoplazmi, kao ribosomska komponenta, ali je takođe otkriven u jedru. Studije na pacovima pokazuju da se ribosomni protein S27 nalazi u blizini ribosomnog proteina S18, u podjedinici 40S i da je kovalentno vezan za faktor inicijacije translacije eIF3. Kao što je tipski za gene koji kodiraju ribosomske proteine, postoji više prerađenih pseudogena ovog gena dispergiranih širom genoma.[5]
Klinički značaj[uredi | uredi izvor]
Njegova ekspresija povećana je kod nekoliko tipova malignosti, a zabilježeno je da nivoi MPS-a opadaju sa liječenjem nekih karcinoma. Također je korišćen kao meta za neke hemoterapije, koje imaju za cilj da helirajući izvuku cink iz motiva cinkovog prsta MPS-a, čineći ga neaktivnim. Ove terapije su se pokazale kao obećavajuće za liječenje raka u laboratorijskim eksperimentima i nekim ograničenim kliničkim ispitivanjima. Karcinom glave i vrata koji je transficiran radi prekomjerne ekspresije ovog proteina pokazao je potisnut rast.[6]
