Protrombin predstavlja plazma protein sa molekulskom težinom od oko 69.000 i normalnom fiziološkom koncentracijom u plazmi od 15 mg/dl. Sintetiše se u jetri gdje je za njegovo normalno stvaranje neophodan vitamin K. Pod uticajem aktivatora protrombina dolazi do njegovog cijepanja u manje komponente kao što je trombin.
